Specimen Type: Amniotic Fluid:
15 mL-20 mL of amniotic fluid in two sterile containers (see Comments).

CVS:
At least 20-25 mg chorionic villi collected in a sterile container, containing Hank's balanced salt solution (HBSS) (see Comments).

Skin and POCs:
Skin (0.5-1cm2), amnion (0.1-5cm2), cord (1cm3), or chorionic villi. Ship in a sterile container, containing Hank's Balanced Salt Solution (HBSS) (preferred) or sterile saline (see comments).
Collection Information: The specimen should be:

• collected aseptically in a sterile container with appropriate medium

• remove and isolate the tissue for study as soon as possible (to prevent contamination)

• If fetal skin is unavailable, then amnion, cord or chorionic villi is acceptable.
Requisition: CYTOGENETICS REQUISITION . (must include patient's name, address, Ontario Health Insurance Number, PIN (if applicable), originating location, Doctor's full name and address, test requested, specimen type and pertinent clinical information.

Special Processing: As products of conception are prone to microbial contamination, collect sample as aseptically as possible and send to the laboratory within 24 hours.
Turnaround times are dependent on the success of culture growth.
Comments: Contact the Cytogenetics Lab for aliquots of Hank’s balanced salt solution for skin and POC samples.

The Cytogenetics Lab is staffed from 0700-1700 (Monday-Friday), Ext. 78974 (office), or 75714 (lab).

Karyotyping (if necessary)
Critical Information Required: Amniotic fluid should be taken with enough time to ensure receipt in the laboratory by 1500 h.
CVS should be taken with enough time to ensure receipt in the laboratory by 1000 h.
All other cell types should be received in the laboratory within 24 hours of collection.
Storage & Shipment: Specimens should shipped same day and be stored at 15- 25ºC during transit. If unable to transport immediately, store at 4oC and transport at room temperature (15-25oC) within 24 hours.
