Details for Autoimmune Liver Disease (AILD) Profile

Test Name: Autoimmune Liver Disease (AILD) Profile
Alternate Name(s): Anti Mitochondrial Antibody (AMA)
Anti Smooth Muscle Antibody (ASMA)
Anti Liver/Kidney Microsome Antibody (Anti LKM)
AILD-relevant antinuclear antibodies (ANA2)
Laboratory: Clinical Immunology
Specimen Type: Serum (preferred) from a 5 mL Gold top Vacutainer tube or 6 mL Red top Vacutainer tube

Pediatric:
0-2 years: Red 0.5 Microtainer tube
2-10 years: 2 mL Red top tube
EDTA, heparin or citrated plasma will also be accepted. Other fluids will not be accepted.
Collection Information: Minimum volume of serum required is 0.5 mL.
Requisition: GENERAL LABORATORY REQUISITION
Test Schedule: Batch Analysis

Reference Range: Negative
Effective Date: 2018-06-11
Revised Date: 2019-01-11
Critical Value:
Interpretive Comments: The following IFA patterns are reported:
AMA
ASMA
Anti LKM
Multiple Nuclear dots
Nuclear membrane
F-Actin

Positive IFA patterns will be titred and evaluated by Immunoblot to identify the following specific antibodies:
AMA M2
M2-3E
Sp100
PML
gp100
LKM-1
LC-1
SLA/LP
Ro-52
Special Processing:
Comments: ANA2 will only screen and report for AILD-related ANA patterns (Multiple Nuclear Dot, Nuclear Membrane and F-Actin).

To screen for other ANA patterns, the regular ANA is tested together with AILDP testing initially.

Repeat testing: For adults ANA repeat testing will not be done; AILDP can be repeated every 6 months. For children AILDP/ANA testing can be repeated every 3 months.
Method of Analysis: Immunofluorescence assay (IFA) for screening
Immunoblot for confirmation
Critical Information Required:
Storage & Shipment: The samples can be stored for up to 14 days at temperatures of 2-8oC. For longer storage, store at -20oC.
Avoid freeze/thaw cycles.
